Effects of Astragaloside Ⅳ on Collagen of Myocardial Fibroblasts in Rats

Abstract:

Objective To investigate the dose- and time-effects of astragalosideⅣ(XGA) on collagen of myocardial fibroblasts in rats.Methods The myocardial fibroblasts of rats were separated by collagenase and trypsinase digestive method, and the cell culture systems were established. After XGA administration with different doses and action times in fibroblasts culture systems, the mRNA express levels of collagen, MMP-1, MMP-2, MMP-9, TIMMP-1, and TIMMP-2, AngⅡ, ET were measured with reverse transcription-PCR(RT-PCR) test.Results After XGA administration with different doses and action times, the gel electrophoresis product of RT-PCR in fibroblasts culture systems expressed the mRNA ofⅠ,Ⅲ,Ⅳtype collagen, MMP-1, MMP-2, MMP-9, TIMMP-1, and TIMMP-2, AngⅡ, ET; but the mRNA express levels ofⅠ,Ⅲ,Ⅳtype collagen,TIMMP-1, and TIMMP-2, AngⅡ, ET decreased and the mRNA express levels of MMP-1, MMP-2, and MMP-9 increased compared to those before XGA administration; the mRNA express levels ofⅠ,Ⅲ,Ⅳtype collagen, MMP-1, MMP-2, MMP-9, TIMMP-1, and TIMMP-2, AngⅡ, ET decreased or increased gradually with increasing of doses and prolonging of action times of XGA. The mRNA express levels ofⅠ,Ⅲ,Ⅳtype collagen, TIMMP-1, and TIMMP-2, AngⅡ, ET were negatively related to the doses and action times of XGA(r=-0.637~-0.973, p=0~0.048); and the mRNA express levels of MMP-1, MMP-2, and MMP-9 were positively related to the doses and action times of XGA (r=0.671~0.962, p=0~0.034).Conclusions XGA could markedly reduce the collagen formation of myocardial fibroblasts in rats, and its mechanisms were possibility related with inhibiting of collagen synthesis and increasing of collagen degradation.
