The Studys of the Alcohol Postmortem Redistribution in Dogs and the Stability in Preservative Samples

Abstract:

Objective:1.To establish the methods of direct-injection gas chromatogram and headspace gas chromatogram in detecting alcohol and self-generation alcohol.2.To establish the models of postmortem redistribution in dogs and stability in preservative samples.3.To study the postmortem redistribution of alcohol in dogs and stability of alcohol in preservative samples.Methods:1.The methods of alcohol and self-generated alcohol detection1.1 Direct-injection gas chromatogram:The samples were diluted with internal standard solution and detected by a TRACE GC2000 equipped with FID and PEG column through the direct-injection.Qualitative analysis was based on retention time and quantitative analysis was based on an internal standard method.1.2 Headspace gas chromatogram:The blood samples were diluted with internal standard solution and distilled water in HS-vials.They were detected by a Model Agilent6820 GC equipped with FID and aDB-5 Model column through the automatic headspace sample injector.Qualitative analysis was based on retention time and quantitative analysis was based on an internal standard method.2.Postmortem redistribution of alcohol2.1 Animal model:Six dogs were given an intragastric administration with a drunkenness dose in ten minutes. They were sacrificed by CO_2 after 1.5 hours.2.2 Samples Collection:The dogs were placed at room temperature.Heart blood,cava blood, humor vitreous,bile,liver,spleen,lung,kidney,brain and muscle were collected at 0,1,4,8,12,24,48,72,96,120 hours after death and was analyzed by direct-injection gas chromatogram respectively.3 The stability of the preservative samplesThe alcohol solution was added in the blank blood which were collected in 12 hours after death and the initial concentrations were 20.12、51.73、77.90、142.29、314.05mg/100ml.They were stored in different vessels(plastic bag,tube,syringe)at different temperature(room temperature,4℃,-20℃)and were analyzed in 2、4、7、15、30 days by the headspace gas chromatogram.4 The study of the self-generated alcohol in stored samples5 Statistics method:The dates was handled by software SPSS11.5 and the result was expressed by x±s.Results:1 The study had established the methods of direct-injection gas chromatogram and headspace gas chromatogram in detecting alcohol and self-generation.The linear range was 2~400mg/100ml,the minimum detectable concentration was 0.5mg/100ml,and the average of recovery rate exceeded 95%2 The study of the post redistribution:2.1 Distribution after death:The sequence of alcohol concentration in dogs was as follows: humor vitreous>left ventricle,left atrium,right ventricle,right atrium,cava,urine>liver, spleen,lung,kidney,cerebrum>bile>muscle after death.The concentration of alcohol of in humor vitreous was the highest which was the 1.4 times of heart blood and cava blood.The ratio of alcohol concentration in bile with heart-blood and cava was 0.67.2.2 Postmortem redistribution:There was a phenomenon of post redistribution in dogs.The left heart blood was more influenced than the humor vitreous,.muscle and cerebrum It should be positive correlation in alcohol and n-propyl alcohol.3 Stability in preservative samples:The concentration alcohol in samples were influenced by the temperature,time,vessel and initial concentration.The interaction was not be observed between temperature and time.4 Self-generated alcohol in stored samples4.1 Self-generated alcohol in stored human blood samples:There was a little alcohol in samples on 30 days at 4℃and no alcohol on 30 days at -20℃.The most samples at room temperature were found a low concentration of alcohol、isopropanol、isobutanol、acetone.4.2 Self-generated alcohol in dogs:There were a different degree of alcohol and n-propyl alcohol concentration in heart blood,cava blood,urine,bile,spleen,lung and kidney,the alcohol concentration in urine was hit 104.92mg/100ml.The cerebrum,humor vitreous and muscle did not easy to corruption.There were a low concentration in brains at 96 hours and in humor vitreous at 72 hoursConclusion:1.The study had established the methods of direct-injection gas chromatogram and headspace gas chromatogram in detecting alcohol and self-generation which was good for separating alcohol and Self-generated alcohol.The methods which had a high sensitivity and could be used in medicolegal identification and study. 2.The concentration of alcohol in dogs as follows:humor vitreous>left ventricle,left atrium,right ventricle,right atrium,cava,urine>liver,spleen,lung,kidney,cerebrum>bile>muscle at the death time.The concentration of alcohol in humor vitreous was the highest which was the 1.4 times of cardiac blood and cava blood.The ratio of alcohol concentration was 0.67 between bile and cardiac blood、cava.3 Postmortem redistribution in dogs.The left heart blood was influenced more seriously by the redistribution than the humor vitreous、muscle and cerebrum.There was a positive correlation between alcohol and n-propyl alcohol.Alcohol,n-propyl alcohol,acetone and isopropanol were generated from the corpse and there was an positive correlation between alcohol and n-propyl alcohol.We should consider the influence of the post interval time for the alcohol concentration in the cases of alcohol poisoned.4 The blood samples were affected by temperature,vessel and initial alcohol concentration.All factors had interaction except of temperature and time.5 there were self-generated alcohol in corrupted human blood and dogs.And there was an positive correlation between alcohol and n-propyl alcohol.The cerebrum,humor vitreous and muscle did not easy to corruption.The alcohol and n-propyl alcohol were found in urine at 12 hours;Alcohol did not detected on 30 days at -20℃.we should not ignore the self-generated alcohol in alcohol cases.
