Studies on Shoot-tips in Vitro Culture and Plant Regeneration of Plum

Abstract:

European plum(Prunus domestica.\’Tardicots\’) and myrobalan(Prunus cerasifera) were used as the materials in the study.By testing the optimal plant growth regulators, their combinations and basal medium combinations,the regeneration system was established using the shoot-tips as explants.The main results were as follows:Firstly,the optimal medium for shoot-tips culture of european plum was MS 6-BA 0.5 mg/L IBA 0.05 mg/L,by 85.5%and 64.16%of seedling rate with 0.35 mm~0.45 mm of shoot-tips and 0.1 mm~0.2 mm of shoot-tips respectively.Secondly,the optimal medium for shoot-tips culture of myrobalan was LP 6-BA 0.5 mg/L IBA 0.05 mg/L,by 84.7%and 64.53%of seedling rate with 0.35 mm~0.45 mm of shoot-tips and 0.1 mm~0.2 mm of shoot-tips respectively.Thirdly,the result of plant subculture and elongation showed that 0.03 mg/L of GA_3 was the best for the elongation and subculture of european plum,0.03 mg/L of GA_3 was the best result for subculture,and 0.05 mg/L of GA_3 was the best for elongation of myrobalan.Fourthly,the medium of shoot rooting was 1/2 MS IBA 1.0 mg/L.At last,to acclimate the seedling,the roots was washed by running water and dipped in the water for 3 days,then the seedlings were transplanted in mixture made of soil,turf and vermiculite(2:1).
